Affinity Analysis Between Molecules

Biolayer interferometry (BLI) is a non-labeling technique based on the principle of optical interference. It has the advantages of simple operation, short detection time, low sample consumption, and direct participation in analysis and detection without manual labeling. By monitoring the optical interference signal in real time, BLI technique can be widely used in the analysis and rapid detection of biomolecular interactions. CD BioSciences provide affinity analysis between molecules service to help customers complete biofilm related research.

Specific Affinity Analysis Between Molecules Approaches We Offer

  • The biosensor is first immersed in a buffer solution for equilibration
  • Immerse in a cured solution of known concentration, the biotinylated antigen in the solution binds to the surface of the biosensor, increasing its surface film thickness
  • Immerse the cured sensor with known concentration of antigen into the buffer solution for baseline
  • Immersion of the cured biosensor with known concentration of antigen into the sample solution containing the antibody to be tested, resulting in an increase in the thickness of the film due to specific binding between the antigen and the antibody
  • The sensor bound with the antibody to be tested was immersed in the buffer solution for dissociation, and the antibody to be tested was shed from the surface of the biosensor resulting in a decrease in the thickness of the membrane. The kinetic constants of the sample to be tested can be obtained by monitoring the biofilm thickness of the biosensor in real time during the experiment.

Selection of Biosensors

When BLI is applied to kinetic constant determination experiments, the commonly used biosensors mainly include SA, SSA, AR2G, APS, AHC and AMC, etc. Each biosensor is suitable for different biological samples, and the requirements for using different biosensors are different. For example, SSA is only suitable for detecting small molecules, while SA is used to solidify biotinylated proteins, antibodies, compounds and nucleic acids to detect large molecules interacting with them, and AHC can directly solidify human antibodies to detect the molecules interacting with them.
