  • TOC quantification of biofilms from CD BioSciences is typically performed in two steps, including the measurement of total carbon (TC) and IC and use to determine TOC. Biofilms are broken down and ICs are converted to CO2, usually acidified by heating and detected by infrared spectroscopy. Next, all carbon in the sample is converted to CO2, usually by thermal oxidation, and TC is then measured. The TOC is then inferred from the difference between these two values ​​(TOC = TC – IC).
  • We use instruments such as carbon analyzers to determine accurate methods for sample preparation and quantification. However, the final calculation of TOC is generic and does not depend on the instrument. Because TOC measures the carbon content of the entire biofilm including bacteria and extracellular polymers (EPS).
  • Although TOC provides a marker to quantify the amount of biofilm present, we nevertheless correlated this value using direct analytical methods (CFU, cell count, etc.) to generate method-independent values. Furthermore, it has been shown that the amount of carbon in a given volume of cells depends on the health of the bacteria. Therefore, we perform additional associations under each set of conditions and must perform a new standardized protocol or control each time an experiment is performed.
