CD BioSciences established a root tip biofilm model through in vitro experiments, and observed the morphology and structure of the Enterococcus faecalis biofilm model.

  • SEM observation

The tissue samples in each group were fixed in glutaraldehyde solution, then dehydrated in graded ethanol, critical point dried, sprayed with gold on the surface of the samples, and observed under SEM.

  • CLSM observation

CLSM observed biofilm layer-by-layer images and overlay images, respectively. Under CLSM, bacteria were stained red by PI, and extracellular matrix was stained green by ConA-FITC.

  • Image analysis

Photoshop CS5 software was used to calculate the biofilm coverage in the SEM field image magnified at 3 000 times, and the biofilm coverage area on the surface of the CLSM sample was calculated with Leica Q Win software.

  • Statistical analysis

SPSS statistical software was used for analysis. The differences in the surface biofilm coverage of SEM observation samples were analyzed by one-way ANOVA. The comparison of means between groups was performed by t test, and the test level was two-sided α=0.05.
