We inoculated the standard and clinical strains of P. acnes with anaerobic broth respectively to adjust the bacterial concentration by McFarland turbidimetric method and inoculated into a 96-well plate. The XTT method was used to detect the absorbance at 490nm and the average value was used to determine the optimal concentration of film-forming bacteria; the bacterial solution was inoculated. 6-well plates with coverslips were placed into 6-well plates for anaerobic culture at different time points for scanning electron microscope (SEM) observation.
