Biofilm Drug Resistance Gene Test Approaches We Offer

  • Drug resistance analysis
    The resistance curve was determined using the standard Kirby-Bauer disk diffusion method according to Clinical and Laboratory Standards Institute (CSLI) guidelines.
  • Biofilm formation test
    We analysed biofilm formation with isolates grown under aerobic conditions in Luria Bertani (LB) agar (Condalab) for 24 hours at 37°C to obtain single colonies. Test the biofilm formation assay in 96-well microtiter plates using the appropriate medium, all plates including sterile controls (medium without inoculum) and growth controls (control medium with inoculum). To avoid evaporation, all plates were covered with adhesive foil lids.
  • Biofilm quantification
    After incubation, cultures were washed with PBS and dried to complete dryness at 65°C. Biofilms were stained with crystal violet (CV) dye solution and incubated for 10 min at room temperature. Measure the optical density (OD) using a microplate reader.
  • Biofilm classification
    In this study, the heterogeneity of sample biomass required defining a cut-off value to classify samples into non-adherent, weakly adherent, moderately adherent, and strongly adherent. For this reason, all samples were tested in triplicate and OD averages were calculated using a negative control (medium without inoculum).
  • Statistical Analysis
    Chi-square test and Spearman's rank correlation test by SPSS were used to study the association and correlation between biofilm formation and antimicrobial susceptibility categories and the respective sources of microorganisms.
